Several experimental approaches to the study of ion channels and the ionic currents they mediate have been developed and applied through the years. However, the control of voltage and simultaneous recording of ionic current, known as voltage clamp, is the most widely deployed. This tool consists on the use of metal electrodes with access to both intracellular and extracellular compartments of a cell or cell-derived membrane. This configuration allows for precise control of membrane potential and high-resolution temporal recording and analysis of the activity of ion channels. Application of voltage-clamp techniques has opened the door to the modern understanding of the biology of channels and other membrane transport proteins. Many of the insights derived from voltage-clamp experiments have been possible with the simultaneous development of exogenous protein expression systems and the application of quantitative analysis methods and, in modern times, in combination with molecular biology and structural biology methods.
